چکیده
Abstract Parkinson’s disease (PD) is the second-most common neurodegenerative worldwide. Several studies have investigated PD for decades; however, exact mechanism of development remains unknown. To study PD, SH-SY5Y cells are often treated with 6-hydroxydopamine (6-OHDA) or 1-methyl-4-phenylpyridinium (MPP+) to induce PD. understand pathogenesis, we confirmed protein changes between 6-OHDA- and MPP+-treated via proteomics analysis using liquid chromatography coupled tandem mass spectrometry. 6-OHDA-treated showed increased expression electron transporter-related proteins compared that in control group, along decreased cells. However, both down- upregulation mitochondrial dysfunction apoptosis. These were protein–protein interaction network IPA STRING Cell-based experiments flow cytometry verified apoptosis membrane potential Our results provide new insights into thereby contributing understanding mechanisms development.

The neuroprotective effect of BSA-based nanocurcumin against 6-OHDA-induced cell death in SH-SY5Y cells
Objective: Parkinson’s disease (PD) is regarded as the second most common neurodegenerative disease affecting elderly population. There is a tendency toward finding natural cures to suppress the initiation and progression of this disease. Some epidemiological studies indicated lower incidence of PD in populations that consume curry. Curcumin, as the main ingredient of turmeric, has been suppose...

Background and Objective: Rheum turkestanicum (R. turkestanicum) has been known to reduce inflammation and has antioxidant properties such as protective effect in neurons. This study aimed to determine the effects of R. turkestanicum on neuronal toxicity induced by the pro-parkinsonian neurotoxin 6-hydroxydopamine (6-OHDA) in neuroblastoma SH-SY5Y cells. Materials and Methods: MTT and DNA frag...
متن کاملProtective effect of (+/-) isoborneol against 6-OHDA-induced apoptosis in SH-SY5Y cells.
Oxidative stress caused by dopamine (DA) may play an important role in the pathogenesis of Parkinson's disease (PD). (+/-) Isoborneol is a monoterpenoid alcohol present in the essential oils of numerous medicinal plants and is a known antioxidant. In this study, we investigated the neuroprotective effect of isoborneol against 6-hydroxydopamine (6-OHDA)-induced cell death in human neuroblastoma ...
متن کاملAlpha-synuclein knockdown attenuates MPP+ induced mitochondrial dysfunction of SH-SY5Y cells.
Alpha-synuclein is one of the main constituents of Lewy bodies and plays an important role in the pathology of Parkinson's disease. Mutation or overexpression of alpha-synuclein causes Parkinson's disease, and downregulation of alpha-synuclein resists MPP(+)-induced cell death, but the mechanism remains elusive. In this study, we attempted to explore the effect of alpha-synuclein knockdown on m...
متن کاملMitochondrial Effects of PGC-1alpha Silencing in MPP+ Treated Human SH-SY5Y Neuroblastoma Cells
The dopaminergic neuron degeneration and loss that occurs in Parkinson's disease (PD) has been tightly linked to mitochondrial dysfunction. Although the aged-related cause of the mitochondrial defect observed in PD patients remains unclear, nuclear genes are of potential importance to mitochondrial function. Human peroxisome proliferator-activated receptor γ coactivator-1alpha (PGC-1α) is a mul...
